How to measure thin flim thickness? Possible using SEM/EDS? If yes,how to do it?
I've tried with profiler and optical microscope using contact-mode but my material is too soft and it got scratched once touched. I coated my material on glass as well as silicon substrates.
What is the nature of your film? If it's transparent, or even semi-transparent (and thin enough), you might be able to use ellipsometry, or perhaps just interferometry.

How does an enzyme recognize its substrate, given that some substrates have optical isomers?
Enzymes will be specific to the form present -Eg.
AMINO ACIDS:-
D and L. While L-amino acids represent the vast majority of amino acids found in proteins, D-amino acids are found in some proteins produced by exotic sea-dwelling organisms, such as cone snails.They are also abundant components of the peptidoglycan cell walls of bacteria. and D-serine may act as a neurotransmitter in the brain. The L and D convention for amino acid configuration refers not to the optical activity of the amino acid itself, but rather to the optical activity of the isomer of glyceraldehyde from which that amino acid can theoretically be synthesized (D-glyceraldehyde is dextrorotary; L-glyceraldehyde is levorotary).
GLUCOSE:- Amylase recognises alpha 1-4 glycosidic bonds and cellulase recognises beta 1-4 glycosidic bonds.
